Baggrund for projektet
Pharmaceutical suspensions can be used for a broad range of applications (e.g., formulation platform for long-acting injectables) associated with several advantages such as simple formulation composition and manufacturing, high drug loading, and the ability to control the release of an active pharmaceutical compound by controlling the sizes of drug particles by different size reduction techniques as for instance, media milling. During milling, the drug compound is suspended in an aqueous dispersion medium containing surfactants and/or polymers for stabilization purposes. Typical stabilizers include polysorbates or poloxamers, where poloxamers are known as amphiphilic triblock copolymers. However, poloxamers have a highly temperature sensitive critical micelle concentration (CMC) as the CMC lowers at increasing temperature. This can greatly alter the behavior and physical stability of the prepared suspension, making it of interest to systematically explore different preparation and storage temperatures during the preparation.
The project will therefore work with three different poloxamers (poloxamer 188, poloxamer 338, and poloxamer 407) in different concentrations to prepare suspensions by media milling and follow the behavior and physical stability as a result of different milling temperatures and subsequent storage temperatures.

Metoder
Suspensions will be prepared using media milling by dual centrifugation and characterized by laser diffraction for stability studies.
All data generated are physical-chemical in nature, hence not associated with any special ethical concerns. All raw data are collected and organized in an easy-to-understand way – and handed over to the supervisor at the end of the project.
